Cultures were initiated from multiple ascospores. A variety of media were utilized, but 2% Difco Potato Dextrose Agar with 5g/L yeast extract (PDYA) was the most favorable for producing stromata. Brightfield microscopy (BF) was accomplished on material mounted in water or Melzer's iodine reagent. Differential interference contrast microscopy (DIC) was done on material mounted in water. Cytological observations were made on nuclei stained with Hoechst 33258 dye (bisbenzimide) following the procedure of Mogford (1979) and examined by fluorescence microscopy (FM) while employing Zeiss exciter filter UG1 and barrier filter 41. Scanning electron microscopy (SEM) was done on material prepared as described by Stahl et al. (1988). Transmission electron microscopy (TEM) utilized material that was fixed in Karnovsky's fixative, sectioned with a diamond knife, and post-section-stained with uranyl acetate and lead citrate (see Stahl et al., 1988 and refs. therein).
